Linperlisib + obinutuzumab is a combination therapy under investigation for the treatment of B-cell lymphomas, including marginal zone lymphoma and follicular lymphoma. Linperlisib is an orally active small molecule inhibitor that selectively targets the delta isoform of phosphatidylinositol 3-kinase (PI3Kδ), which plays a key role in B-cell receptor signaling and survival of malignant B-cells[1][3]. Obinutuzumab is a fully humanized, glycoengineered type II anti-CD20 monoclonal antibody designed to enhance B-cell killing through mechanisms such as direct induction of apoptosis, antibody-dependent cellular cytotoxicity (ADCC), phagocytosis, and complement activation[6][8][10]. The combination aims to leverage complementary mechanisms—targeted inhibition of PI3Kδ-mediated survival pathways by linperlisib and potent depletion of CD20-positive B-cells by obinutuzumab—to improve outcomes in patients with relapsed or refractory lymphomas. Clinical trials are ongoing to evaluate efficacy and safety in marginal zone lymphoma[4].
