Drug intelligence / Profile preview
This is a **combination regimen** of **linperlisib**, a next-generation, highly selective, oral **PI3K delta (PI3Kδ) inhibitor**, and **rituximab**, a monoclonal antibody targeting **CD20** on B lymphocytes. Linperlisib works by selectively inhibiting the PI3Kδ isoform, a key signaling molecule in B-cell receptor signaling pathways, thereby reducing proliferation and survival of malignant B-cells. Rituximab binds to CD20, leading to B-cell lysis via immune-mediated mechanisms. This combination is being explored in the treatment of **relapsed or refractory indolent B-cell lymphomas** and other B-cell malignancies, as well as in **relapsed/refractory large B-cell lymphoma (LBCL)** and related conditions. The combination aims to leverage the targeted cytotoxicity of rituximab with the pathway-specific inhibition of linperlisib for enhanced antitumor efficacy[3][5][7][10].
