Lintuzumab satetraxetan AC‑225 is a radioimmunoconjugate composed of the humanized anti-CD33 monoclonal antibody, lintuzumab, conjugated to the alpha-emitting radioisotope actinium‑225. This drug targets CD33, a cell surface antigen expressed on most acute myeloid leukemia (AML) cells and some other myeloid malignancies. Upon binding to CD33-positive cells, the attached actinium‑225 delivers potent alpha-particle radiation directly to malignant cells, causing double-stranded DNA breaks and cell death with minimal impact on surrounding healthy tissue due to the short path length of alpha emissions. Lintuzumab satetraxetan AC‑225 is being developed primarily for AML and high-risk myelodysplastic syndromes (MDS), with ongoing research in other cancers expressing CD33. The drug has orphan designation for AML and is under development by Actinium Pharmaceuticals in collaboration with several academic institutions[1][2][5][6][7].
