Lio-Msln TCR-T is an autologous T-cell receptor (TCR) engineered T-cell therapy being developed by Lion TCR for the treatment of mesothelin-expressing solid tumors. The therapy involves genetically modifying a patient's own T cells to express a TCR specific to mesothelin (MSLN), a protein that is overexpressed in various malignancies including ovarian and pancreatic cancers but shows minimal expression in healthy tissues. Upon administration, these engineered T cells recognize mesothelin peptides presented by MHC molecules on tumor cells, leading to targeted cell lysis. Research has indicated that combining Lio-Msln TCR-T with checkpoint inhibitors (such as anti-PD-1, anti-Lag-3, and anti-Tim-3) may enhance anti-tumor efficacy by mitigating the immunosuppressive tumor microenvironment.
