Overview

LioCyx-M is an investigational autologous cell therapy developed by Lion TCR for the treatment of hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC). It consists of a preparation of autologous patient-derived T cells that are transfected with mRNA encoding a hepatitis B surface antigen (HBsAg)-specific T cell receptor (TCR), redirecting the immune response specifically against HBV-infected liver cancer cells. The therapy aims to directly lyse HCC cells expressing HBV antigens and modulate the tumor microenvironment, potentially converting immunologically "cold" tumors into "hot" ones characterized by increased proinflammatory cytokine production and enhanced immune infiltration. Clinical trials have shown that LioCyx-M infusions are generally well tolerated, with no cytokine release syndrome or neurotoxicity observed, and some patients have experienced durable tumor responses. The product is being evaluated as monotherapy and in combination with lenvatinib in global Phase 2 clinical trials[1][3][6].
