Drug intelligence / Profile preview
Lip-Con is a cationic liposomal formulation of trehalose 6,6'-dimycolate (TDM) purified from the Mycobacterium bovis BCG Connaught strain. Developed as a non-living, safer alternative to intravesical BCG therapy for bladder cancer, Lip-Con functions as a potent agonist of the Macrophage-inducible C-type lectin (Mincle) receptor. Upon administration, it activates Mincle on dendritic cells within the tumor microenvironment, promoting their maturation and migration to tumor-draining lymph nodes. This process triggers a robust CD8+ T-cell-mediated antitumor immune response. Preclinical studies have demonstrated that Lip-Con exhibits antitumor efficacy comparable or superior to live BCG in models of bladder cancer, colon cancer, and melanoma, without the associated risks of systemic infection, body weight loss, or granuloma formation.
