LIPO-HSV-1-tk is a liposomal gene therapy formulation that delivers the herpes simplex virus type 1 thymidine kinase (HSV-1-tk) gene directly to target tissues, typically via intratumoral infusion. The HSV-1-thymidine kinase enzyme phosphorylates certain nucleoside analogues (such as ganciclovir or acyclovir), converting them into toxic metabolites that induce cell death. This "suicide gene" approach is used in cancer therapy to selectively kill tumor cells transduced with the HSV-1-tk gene when patients are subsequently treated with these prodrugs. The use of a liposome-based delivery system enhances cellular uptake and localizes the effect to the tumor site[1][2].
