Lipoic acid trisulfide (LATS) is a small molecule derivative of lipoic acid containing a trisulfide bridge, designed to function as a hydrogen sulfide (H2S) donor. It serves as the monomeric precursor for the polymeric nanomedicine poly(lipoic acid trisulfide) (pLATS). LATS undergoes thiol-triggered intracellular exchange and depolymerization to release H2S and lipoic acid (LA), which together provide synergistic antihypertensive and renoprotective effects. The molecule modulates the renin-angiotensin system and improves endothelial function, while also upregulating endogenous H2S-producing enzymes to promote sustained H2S biogenesis. It is primarily investigated for the treatment of hypertensive chronic kidney disease (CKD), where it has demonstrated the ability to lower blood pressure and improve renal biomarkers such as the urine albumin-to-creatinine ratio.
