LiPoNs (Lipid-Polymer Nanoparticles) are hybrid nanostructures engineered for the simultaneous delivery of therapeutic and diagnostic agents, specifically targeting triple-negative breast cancer (TNBC). These nanoparticles are synthesized using a microfluidic platform called coupled Hydrodynamic Flow Focusing (cHFF), which allows for precise control over the self-assembly of lipid and polymer (chitosan) components. The resulting hybrid architecture combines the biocompatibility and membrane fusion capabilities of lipids with the structural robustness and controlled release properties of polymers. In the context of TNBC theranostics, LiPoNs are used to co-deliver AntimiR-21, an antisense oligonucleotide that suppresses the oncogenic microRNA-21, and gadolinium diethylenetriamine penta-acetic acid (Gd-DTPA), a contrast agent for magnetic resonance imaging (MRI).
