Lipophosphonoxins (LPPOs) are a novel class of small-molecule antimicrobial agents developed by IOCB TECH, the technology transfer arm of the Institute of Organic Chemistry and Biochemistry of the Czech Academy of Sciences. These compounds act through a unique mechanism of action by targeting and disrupting the bacterial cytoplasmic membrane, leading to rapid depolarization and cell death. LPPOs exhibit broad-spectrum activity against both Gram-positive and Gram-negative bacteria, including multi-drug resistant strains such as methicillin-resistant *Staphylococcus aureus* (MRSA) and vancomycin-resistant enterococci (VRE). The modular structure of LPPOs, typically consisting of a nucleoside-derived scaffold linked to a lipophilic chain via a phosphonate group, allows for optimization for various applications, including topical treatments for skin and soft tissue infections and potential systemic administration.
