Liposomal alendronate is a formulation of the bisphosphonate drug alendronate encapsulated within liposomes to improve its pharmacokinetic properties and therapeutic profile. Alendronate works by inhibiting bone resorption through its action on osteoclasts, thereby increasing bone density. The liposomal formulation enhances the stability and circulation time of alendronate in the bloodstream, reduces premature drug leakage, and allows for targeted delivery to specific tissues such as bone or tumors. This approach aims to increase bioavailability, reduce gastrointestinal side effects associated with oral administration, and enable new therapeutic applications such as immunomodulation in cancer therapy by targeting macrophages and other myeloid cells. Liposomal alendronate has been investigated for use in osteoporosis, Paget's disease of bone, prevention of corticosteroid-induced osteoporosis, restenosis after vascular stenting (by depleting monocytes/macrophages), and as an immunotherapeutic agent in oncology.
