Overview

Liposomal amphotericin B is a lipid-based formulation of the antifungal agent amphotericin B. It is designed to reduce the toxicity associated with conventional amphotericin B by encapsulating the drug in unilamellar liposomes composed of phospholipids and cholesterol. This formulation allows for targeted delivery to infected tissues and reduced nephrotoxicity compared to conventional formulations. Liposomal amphotericin B acts by binding to ergosterol, a key component of fungal cell membranes, forming pores that increase membrane permeability and lead to cell death. It is primarily indicated for serious systemic fungal infections (such as invasive aspergillosis, candidiasis, cryptococcal meningitis) and visceral leishmaniasis, especially in patients who cannot tolerate or have failed standard therapy[1][2][5][6][7].
