Liposomal annamycin plus cytarabine is an investigational combination therapy for the treatment of acute myeloid leukemia (AML), particularly in patients with relapsed or refractory disease. Liposomal annamycin (L-ANN) is a novel, liposome-encapsulated anthracycline designed to overcome multidrug resistance and reduce cardiotoxicity compared to traditional anthracyclines like doxorubicin. It acts as a **topoisomerase II alpha** poison, inducing apoptosis in cancer cells. Cytarabine is a nucleoside analog that inhibits **DNA synthesis** and is widely used in AML treatment regimens. The combination has shown promising safety and efficacy results in early-phase clinical trials, with no significant cardiotoxicity observed and notable response rates as second-line therapy for AML patients who lack targetable mutations such as FLT3 or IDH1/2[1][5][7][8].
