Drug intelligence / Profile preview
Liposomal C6 ceramide is a nanoliposomal formulation of the bioactive sphingolipid C6 ceramide (N-hexanoyl-D-erythro-sphingosine) designed to enhance the intracellular delivery and therapeutic efficacy of ceramide. Ceramide acts as a pro-apoptotic signaling molecule that is often downregulated in cancer cells. By delivering exogenous C6 ceramide, the compound inhibits the Akt/mTOR signaling pathway through the activation of protein phosphatase 1 (PP1) and induces caspase-dependent apoptosis. Developed primarily in the laboratory of Mark Kester, it has shown preclinical activity in various malignancies, including acute myelogenous leukemia (AML), melanoma, and solid tumors, often demonstrating synergy with other agents that modulate sphingolipid metabolism.
