Overview
**Liposomal curcumin + temozolomide** is a combination therapy under clinical investigation for high-grade gliomas, especially glioblastoma. Liposomal curcumin is a nanoparticle-formulated version of curcumin designed to enhance bioavailability and delivery to tumor tissue, while temozolomide is an oral alkylating chemotherapy agent. Curcumin modulates multiple signaling pathways in cancer cells (e.g., cell proliferation, apoptosis, cell cycle, autophagy, oxidative stress), and has been shown to sensitize glioblastoma cells to temozolomide, promoting synergistic effects. Temozolomide induces cytotoxicity primarily by methylating DNA at the O6 and N7 positions of guanine, leading to DNA damage and apoptosis in tumor cells.
