Liposomal doxorubicin + cisplatin is a combination chemotherapy regimen consisting of pegylated liposomal doxorubicin (PLD), a liposome-encapsulated form of doxorubicin with prolonged circulation time, and cisplatin, a platinum-based DNA cross-linking agent. PLD works by inhibiting topoisomerase II and generating free radicals that damage DNA, delivered in liposomal form to enhance tumor targeting and reduce cardiotoxicity. Cisplatin forms DNA crosslinks, disrupting DNA synthesis and inducing apoptosis. This combination is investigated or used for cancers such as platinum-sensitive ovarian cancer and malignant pleural mesothelioma, with phase II evidence showing promising antitumor activity and manageable toxicity.
