Drug intelligence / Profile preview
Liposomal eribulin (E7389-LF) is a liposome-encapsulated formulation of the microtubule inhibitor eribulin, designed to enhance tumor delivery and alter pharmacokinetics relative to conventional eribulin mesylate. Eribulin is a synthetic analog of the marine natural product halichondrin B that binds tubulin, suppresses microtubule growth, induces G2/M cell-cycle arrest, and triggers apoptotic cell death; in addition, eribulin has been shown to promote tumor vascular remodeling and may modify the tumor microenvironment. Early-phase studies of liposomal eribulin have evaluated intravenous dosing every 3 weeks in patients with advanced solid tumors, including gastric, esophageal, adenoid cystic, and small cell lung cancers, both as monotherapy and in combination with the PD-1 antibody nivolumab, demonstrating manageable safety and preliminary antitumor activity.
