Liposomal interleukin-2 is a formulation in which recombinant human interleukin-2 (IL-2), a cytokine that stimulates the proliferation and activation of T cells and natural killer (NK) cells, is encapsulated within lipid-based vesicles (liposomes). This delivery system enhances the localization and retention of IL-2 at tumor sites while reducing systemic toxicity compared to free or pegylated IL-2. Liposomal encapsulation allows for more targeted immune activation within the tumor microenvironment, leading to increased infiltration by cytotoxic lymphocytes and improved antitumor responses. Preclinical studies have demonstrated that localized or aerosolized delivery of liposomal IL-2 can suppress tumor growth in various animal models—including pulmonary metastases—while minimizing adverse effects commonly associated with high-dose systemic IL-2 therapy. Liposomal formulations are also being explored in combination with chemotherapy or other immunotherapies to further enhance efficacy[7][8][2].
