Drug intelligence / Profile preview
Liposomal interleukin-2 (L-IL-2) is a lipid-encapsulated formulation of recombinant interleukin-2 (aldesleukin) designed to improve the therapeutic index of IL-2 therapy. Interleukin-2 is a potent cytokine that stimulates the proliferation and activation of T cells and natural killer (NK) cells, which are critical for anti-tumor immunity. However, high-dose systemic administration of free IL-2 is associated with severe toxicities, most notably vascular leak syndrome (VLS). By encapsulating the cytokine in liposomes, L-IL-2 aims to alter the pharmacokinetics and biodistribution of the drug, potentially reducing systemic toxicity while maintaining or enhancing anti-tumor efficacy through sustained release and improved delivery to lymphoid tissues and tumor sites. It has been investigated primarily for the treatment of metastatic melanoma and renal cell carcinoma.
