This combination chemo-immunotherapy regimen is being investigated for the treatment of extensive-stage small cell lung cancer (ES-SCLC). The regimen consists of an induction phase utilizing liposomal irinotecan (a nanoparticle-encapsulated topoisomerase I inhibitor), a platinum agent (cisplatin or carboplatin), the anti-PD-1 monoclonal antibody tislelizumab, and the multi-targeted tyrosine kinase inhibitor anlotinib. This is followed by a maintenance phase of tislelizumab and anlotinib. Liposomal irinotecan is designed to provide sustained drug release and enhanced intratumoral accumulation compared to conventional irinotecan. Tislelizumab restores anti-tumor immune responses by blocking the PD-1/PD-L1 pathway, while anlotinib inhibits tumor angiogenesis and proliferation by targeting VEGFR, FGFR, PDGFR, and c-Kit. The regimen is currently being evaluated in Phase II clinical trials sponsored by China Medical University.
