Liposomal mitoxantrone hydrochloride + pegaspargase is a combination therapy under investigation primarily for the treatment of relapsed or refractory peripheral T-cell lymphoma and extranodal NK/T-cell lymphoma. Liposomal mitoxantrone is a formulation of the anthracenedione antineoplastic agent, encapsulated in a lipid bilayer to improve pharmacokinetics and reduce toxicity. It acts as a DNA intercalator and type II DNA topoisomerase inhibitor, disrupting DNA synthesis and repair in cancer cells[6][8]. Pegaspargase is an enzyme that depletes asparagine by converting it to aspartic acid, exploiting the inability of certain leukemia/lymphoma cells to synthesize asparagine, leading to cell death[2][5]. The combination aims to enhance efficacy while minimizing side effects compared with standard regimens.
