Overview

Liposomal NDDP (L-NDDP) is a liposomal formulation of cis-bis-neodecanoato-trans-R,R-1,2-diaminocyclohexane platinum(II), which is a structural analogue of oxaliplatin. The drug is formulated in large multilamellar liposomes (1-3 micrometers in size)[9]. This liposomal encapsulation technology helps protect the active compound from physiological degradation, extends its half-life, and allows for controlled release of the drug molecules[2]. L-NDDP has been investigated primarily for the treatment of advanced colorectal cancer in patients who are refractory to standard treatments like 5-fluorouracil/leucovorin or capecitabine and irinotecan[6][8]. It has also been studied for intrapleural administration in patients with malignant pleural effusions, including those with adenocarcinoma of the lung, malignant pleural mesothelioma, and ovarian carcinoma[9]. The liposomal formulation provides several advantages over conventional drug delivery, including reduced toxicity profiles compared to free platinum compounds, enhanced circulation time, and potential for improved efficacy through targeted delivery to tumor sites[1][2].
