Liposomal paclitaxel + cisplatin + S-1 is a combination chemotherapy regimen used primarily in the treatment of advanced gastric cancer and non-small cell lung cancer. Liposomal paclitaxel (marketed as Lipusu) is a nanoparticle formulation designed to improve the delivery and reduce the toxicity of conventional paclitaxel. Cisplatin, sometimes delivered in a liposomal form (such as Lipoplatin), is a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis in rapidly dividing cells. S-1 is an oral fluoropyrimidine derivative combining tegafur with two modulators to enhance efficacy and reduce gastrointestinal toxicity. The combination leverages multiple mechanisms: microtubule stabilization by paclitaxel, DNA damage by cisplatin, and inhibition of thymidylate synthase by S-1, resulting in synergistic antitumor effects[2][3][6]. This regimen has demonstrated efficacy with manageable toxicity profiles in patients with poor performance status or advanced disease.
