Drug intelligence / Profile preview
Liposomal prednisolone is a novel intravenous formulation of the synthetic glucocorticoid corticosteroid prednisolone. In this formulation, the active drug is encapsulated within long-circulating (PEGylated) liposomes to enhance targeted delivery to inflamed tissues and reduce systemic exposure. This approach aims to improve the therapeutic index of glucocorticoids by increasing efficacy at sites of inflammation while minimizing adverse effects associated with systemic corticosteroid therapy. Liposomal prednisolone acts primarily as an agonist at the glucocorticoid receptor, inhibiting inflammatory pathways such as arachidonic acid metabolism and modulating immune responses. It has been investigated for use in rheumatoid arthritis (Phase III), arteriovenous fistula maturation (Phase II), Graves ophthalmopathy (Phase II), and ulcerative colitis (Phase II). Clinical studies suggest it may offer superior or comparable efficacy with potentially fewer side effects compared to conventional steroid regimens[2][4][5].
