Liposomal ropivacaine hydrochloride (LY09606) is a multivesicular liposome formulation of the long-acting amide local anesthetic ropivacaine hydrochloride. Developed by Luye Pharma, LY09606 is designed for local infiltration to provide sustained-release postoperative analgesia. Ropivacaine acts as a sodium channel blocker, preventing the generation and conduction of nerve impulses to produce local anesthesia and analgesia. The multivesicular liposome structure of LY09606 allows for a slow, controlled release of ropivacaine, extending the duration of action and potentially reducing the need for postoperative opioids or continuous infusion devices while minimizing systemic cardiac and central nervous system toxicities compared to bupivacaine-based liposomal formulations. It is currently in Phase I clinical development in China for postoperative pain management, such as after hemorrhoidectomy.
