Liposomal transcrocetin (LEAF-4L6715) is a liposomal formulation of transcrocetin (trans sodium crocetinate), a bipolar trans-carotenoid salt that acts as an oxygen diffusion enhancer. It is designed to facilitate the movement of oxygen from red blood cells into hypoxic tissues by increasing the diffusivity of oxygen in aqueous solutions like plasma. The liposomal delivery system allows for a gradual release of the active compound, which facilitates less frequent dosing compared to the non-liposomal form. Liposomal transcrocetin is primarily being investigated for the treatment of Acute Respiratory Distress Syndrome (ARDS) associated with COVID-19, sepsis, or other causes, where it aims to improve oxygenation (PaO2/FiO2 ratio). Additionally, it is in clinical development as a radiosensitizer for solid tumors, including glioblastoma and soft tissue sarcoma, by reducing tumor hypoxia and thereby enhancing the efficacy of radiotherapy.
