Drug intelligence / Profile preview
Liposome amphotericin B is a unilamellar (single-layer) liposomal formulation of the antifungal agent amphotericin B. It is designed for intravenous administration and consists of amphotericin B intercalated within a true single bilayer liposome composed of phospholipids and cholesterol. This formulation improves the drug’s safety profile by reducing nephrotoxicity compared to conventional formulations. Liposome amphotericin B acts by binding to ergosterol in fungal cell membranes, creating pores that lead to cell death. It is FDA-approved for empirical therapy in febrile neutropenic patients with presumed fungal infection; treatment of invasive infections caused by Aspergillus species, Candida species, and/or Cryptococcus species refractory to conventional therapy; cryptococcal meningitis in HIV-infected patients; and visceral leishmaniasis[2][3][4][5][9]. Off-label uses include coccidioidomycosis, histoplasmosis, mucormycosis, blastomycosis, talaromycosis[5][6]. The product was developed as an improved delivery system for systemic antifungal therapy.
