Lipotecan is a novel camptothecin analog and topoisomerase I inhibitor developed as an anticancer agent. It is formulated as a liposomal preparation (TLC388) to enhance delivery and efficacy. Lipotecan induces immunogenic cell death, increases cancer immunogenicity, and remodels the tumor microenvironment to provoke antitumor immune responses. Preclinical studies suggest it can enhance the therapeutic response to radiotherapy and immune checkpoint inhibitors by activating STING pathways. Clinical trials have evaluated its use in advanced solid tumors, hepatocellular carcinoma, and poorly differentiated neuroendocrine carcinomas[8][10].
