Overview
Liraglutide + metformin is a combination therapy being investigated for the treatment of obesity and associated metabolic disorders. Liraglutide is a long-acting glucagon-like peptide-1 (GLP-1) receptor agonist that enhances glucose-dependent insulin secretion, suppresses inappropriate glucagon secretion, and slows gastric emptying to promote satiety and weight loss. Metformin is a biguanide that primarily reduces hepatic glucose production and improves peripheral insulin sensitivity, often through the activation of AMP-activated protein kinase (AMPK). This specific combination is being evaluated by Xiangya Hospital of Central South University in a Phase 2 clinical trial to determine its efficacy in weight loss and metabolic improvement in obese Chinese patients, particularly those who do not achieve adequate glycemic control with liraglutide monotherapy. The study also explores pharmacogenomic predictors to identify genetic factors influencing the curative effect of the treatment.
