Overview

This combination therapy regimen consists of lirilumab, nivolumab, and azacitidine, and is primarily investigated for the treatment of myelodysplastic syndromes (MDS). Lirilumab is an investigational human IgG4 monoclonal antibody that blocks killer immunoglobulin-like receptors (KIR) on natural killer (NK) cells, thereby enhancing their anti-tumor activity. Nivolumab is a programmed death-1 (PD-1) checkpoint inhibitor that activates T-cell-mediated immune responses. Azacitidine is a hypomethylating agent that exerts antineoplastic effects by inducing DNA hypomethylation and direct cytotoxicity on abnormal hematopoietic cells. The combination aims to synergistically enhance immune surveillance against MDS cells by simultaneously targeting inhibitory signals on both NK and T cells while utilizing the epigenetic modulating effects of azacitidine.
