Lisaftoclax + rituximab is an investigational combination therapy comprised of lisaftoclax, an oral, highly selective BCL-2 inhibitor, and rituximab, a monoclonal antibody targeting CD20 on B-cells. **Lisaftoclax** inhibits the BCL-2 protein, promoting apoptosis in malignant cells overexpressing BCL-2 (notably chronic lymphocytic leukemia [CLL] and small lymphocytic lymphoma [SLL]), and is currently in late-stage development. **Rituximab** induces depletion of CD20-expressing B-cells through several mechanisms, including complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. This combination is being investigated, particularly in patients with treatment-naïve and relapsed/refractory CLL/SLL, for its synergistic effects and favorable safety profile[1][3][5][7][8].
