Lishatoclax (also known as lisaftoclax or APG-2575) is an orally bioavailable, highly selective small-molecule inhibitor of B-cell lymphoma-2 (BCL-2). It is designed to induce apoptosis in cancer cells by restoring the apoptotic signaling pathway, specifically by displacing pro-apoptotic proteins from the BH3-binding groove of BCL-2. Developed by Ascentage Pharma, it is being investigated for the treatment of various hematologic malignancies, including chronic lymphocytic leukemia (CLL), acute myeloid leukemia (AML), and myelodysplastic neoplasms (MDS). In clinical trials, such as the Phase II study ChiCTR2600125359, it is evaluated in combination with hypomethylating agents like decitabine, particularly in patients who have failed prior therapies such as venetoclax.
