Overview

Lisocabtagene maraleucel is a CD19-directed genetically modified autologous T cell immunotherapy, classified as a chimeric antigen receptor (CAR) T-cell therapy. It is used for the treatment of adults with certain types of B-cell lymphomas and leukemias that are relapsed or refractory to prior therapies. The therapy involves collecting a patient's own T cells, genetically modifying them ex vivo to express a CAR targeting the CD19 antigen on B cells, and then infusing them back into the patient. The CAR construct includes an anti-CD19 single chain variable fragment (scFv), 4-1BB costimulatory domain, and CD3-zeta activation domain. Upon administration, these engineered T cells recognize and selectively kill CD19-expressing malignant B cells[2][5][6][8]. Lisocabtagene maraleucel was developed by Juno Therapeutics (a Bristol Myers Squibb company) and is marketed under the brand name Breyanzi.
