Drug intelligence / Profile preview
Listeria-PV (AWAKE-LM-PV) is a preclinical-stage cancer immunotherapy developed by Loki Therapeutics using their proprietary AWAKE platform. The therapy utilizes a live attenuated *Listeria monocytogenes* bacterial vector to deliver polio vaccine recall antigens directly into the tumor microenvironment and tumor cells. By presenting these antigens, the drug aims to redirect and activate pre-existing memory T cells—originally generated through childhood polio vaccinations—to recognize and eliminate cancer cells. This approach leverages the potent innate and adaptive immune responses triggered by *Listeria* alongside the high frequency of memory T cells specific to common vaccine antigens, bypassing the need for the immune system to generate a *de novo* response against tumor neoantigens.
