Overview

Listeria-TT is an investigational immunotherapy platform developed by Loki Therapeutics in collaboration with the Albert Einstein College of Medicine. It utilizes a live-attenuated strain of *Listeria monocytogenes* as a bacterial vector to deliver the tetanus toxoid (TT) protein directly into tumor cells and the tumor microenvironment. The therapeutic strategy is based on "recall antigen" technology, which leverages the fact that most individuals have been previously vaccinated against tetanus and possess circulating memory T cells specific to the TT protein. By delivering and expressing the TT antigen within the immunosuppressive tumor microenvironment, Listeria-TT reactivates these pre-existing memory T cells to recognize and destroy the infected cancer cells. This approach is primarily being developed for the treatment of advanced solid tumors, with a significant focus on pancreatic cancer.
