Overview

Litifilimab is a humanized IgG1 monoclonal antibody that targets blood dendritic cell antigen 2 (BDCA2), a receptor exclusively expressed on plasmacytoid dendritic cells (pDCs)[5][7]. By binding to BDCA2, litifilimab inhibits the activity of pDCs and reduces the production of type I interferons and other proinflammatory cytokines and chemokines[1][5][6]. These mediators are implicated in the pathogenesis of autoimmune diseases such as systemic lupus erythematosus (SLE) and cutaneous lupus erythematosus (CLE)[3][5]. Litifilimab has demonstrated efficacy in phase II clinical trials for both SLE and CLE by reducing skin disease activity and joint involvement[3][5]. The drug was discovered and developed by Biogen.
