Overview

Litoxetine (also known as IXA-001 or VNX005) is a small molecule drug candidate originally developed by Sanofi as an antidepressant (SL 81.0385). It functions as a selective serotonin reuptake inhibitor (SSRI) with additional antagonist activity at the 5-HT3 receptor. The drug was later licensed to Ixaltis and Vaneltix Pharma for repurposing in the treatment of urological conditions. It is currently in Phase 2 development for stress urinary incontinence (SUI) and mixed urinary incontinence (MUI). The therapeutic rationale for its use in incontinence is based on its ability to increase serotonin concentrations in the Onuf's nucleus of the sacral spinal cord, which in turn increases the contraction of the external urethral sphincter via the pudendal nerve.
