Overview

LEAP2 (Liver-Expressed Antimicrobial Peptide 2) is an endogenous 40-amino acid peptide that acts as a natural antagonist and inverse agonist of the growth hormone secretagogue receptor (GHSR), also known as the ghrelin receptor. Developed as an investigational biologic by the University Hospital, Gentofte, Copenhagen, it is being evaluated in the SILENCED clinical trial for its ability to modulate appetite and metabolic responses in individuals with obesity who are concurrently treated with the GLP-1 receptor agonist semaglutide. By competitively inhibiting ghrelin binding and reducing the constitutive activity of the GHSR, LEAP2 aims to clarify the physiological role of ghrelin signaling during GLP-1-induced appetite suppression and potentially identify new therapeutic pathways for weight management.
