Lixadesiran is a small interfering RNA (siRNA) therapeutic under development for the treatment of cutaneous squamous cell carcinoma (cSCC) and other skin-related conditions. It acts by targeting and inhibiting cyclooxygenase-2 (COX-2), an enzyme involved in inflammation and tumorigenesis, as well as transforming growth factor beta 1 (TGF-β1), which plays a role in fibrosis and cancer progression. Lixadesiran is administered via intradermal injection, topical cream, intra-tumoral, or subcutaneous routes. The drug is being developed using Sirnaomics' proprietary PNP platform and has reached Phase II clinical trials for cSCC. Other indications explored include basal cell carcinoma, primary sclerosing cholangitis, keloid scar healing, hypertrophic scars, abdominal obesity, pulmonary fibrosis, kidney fibrosis, wounds, pancreatic cancer, non-alcoholic steatohepatitis (NASH), cholangiocarcinoma, hepatocellular carcinoma and bladder cancer[2][4][7].
