Overview

LJP 1082 is a small molecule immunosuppressant developed as a "toleragen" for the treatment of antibody-mediated autoimmune diseases, particularly antiphospholipid syndrome (APS) and thrombosis. It was designed as a polyvalent antigenic structure that crosslinks specific surface immunoglobulin on B cells, thereby tolerizing them to beta2-glycoprotein I (β2GPI), the primary autoantigen in APS. By inducing immune tolerance at the B-cell level, LJP 1082 aimed to reduce pathogenic autoantibodies without broadly suppressing healthy immune function. The drug was intended to address limitations of standard anticoagulant therapy by specifically targeting disease-causing antibodies and their related B cells[3][4][5][9]. Development was discontinued after early clinical trials.
