**LL-E7P-IL-12D** is a preclinical intranasal therapeutic cancer-vaccine candidate consisting of a live, cell-wall-weakened recombinant *Lactococcus lactis* strain engineered to express human papillomavirus type 16 E7 protein and to deliver a eukaryotic expression cassette encoding murine interleukin-12. It was designed to generate HPV-16 E7-specific cytotoxic T-cell responses while locally increasing IL-12-driven immune activation. In C57BL/6 mice bearing HPV-16 E7-expressing TC-1 tumors, intranasal administration induced E7-specific cytotoxic lymphocyte activity, increased serum IL-12 and interferon-gamma, inhibited tumor growth, and produced tumor-free animals in prophylactic and therapeutic experiments.
