LL-RP-GD is a recombinant strain of the food-grade bacterium *Lactococcus lactis* NZ3900, engineered to serve as a dual-reporter control vector in mucosal vaccine and gene therapy research. It contains two distinct expression systems: a prokaryotic cassette (P32 promoter) for the constitutive expression of Red Fluorescent Protein (RFP) and a eukaryotic expression cassette for the delivery of enhanced Green Fluorescent Protein (eGFP) DNA to mammalian cells. Developed by researchers at Xinjiang University, LL-RP-GD is primarily utilized as a negative control in studies evaluating therapeutic lactococcal vaccines, such as those targeting HPV-16 E7. It allows researchers to account for the baseline immunogenicity of the *L. lactis* vector and verify the successful co-delivery of both protein and DNA payloads to mucosal surfaces via intranasal administration.
