LLE-aCD38 is an investigational mRNA-based therapeutic developed by Hangzhou LLE Biopharma for the treatment of hematologic malignancies, specifically multiple myeloma. The drug utilizes a proprietary Lipid-Like Envelope (LLE) nanoparticle delivery system to encapsulate mRNA that encodes a full-length monoclonal antibody targeting CD38. Following intravenous administration, the mRNA is taken up by host cells (primarily in the liver) and translated into the anti-CD38 antibody, which is then secreted into the systemic circulation. This in vivo production method aims to maintain stable therapeutic concentrations of the antibody while potentially reducing the frequency of administration and manufacturing costs associated with traditional recombinant protein production. The secreted antibody functions by binding to CD38-expressing malignant plasma cells, triggering tumor cell death through antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and antibody-dependent cellular phagocytosis (ADCP).
