Overview

LLL12B is a novel small molecule inhibitor of Signal Transducer and Activator of Transcription 3 (STAT3). Developed by researchers at the University of Florida, it is primarily being investigated for the treatment of glioblastoma (GBM). LLL12B works by blocking the phosphorylation and activation of STAT3, which is a key regulator of myeloid-derived suppressor cells (MDSCs). By inhibiting STAT3, LLL12B impairs the mobilization and recruitment of MDSCs from the bone marrow to the tumor microenvironment, thereby mitigating immune suppression. Preclinical studies have shown that LLL12B can reduce tumor volume and extend survival in murine GBM models, particularly when used in combination with immune checkpoint inhibitors like anti-PD-1 or focal radiation.
