Drug intelligence / Profile preview
LLO-CD105A (also known as Lm-LLO-CD105A) is a live-attenuated, recombinant *Listeria monocytogenes* (Lm)-based cancer vaccine designed to target endoglin (CD105), a glycoprotein highly expressed on tumor cells and proliferating endothelial cells in tumor-associated neovasculature. The vaccine consists of an attenuated Lm vector engineered to secrete a fusion protein containing a truncated, non-hemolytic fragment of listeriolysin O (LLO) fused to an antigenic fragment of CD105. Upon administration, LLO-CD105A is internalized by antigen-presenting cells, where LLO facilitates phagosomal escape, allowing the CD105 antigen to be processed and presented via MHC class I and II pathways. This stimulates a robust, antigen-specific CD8+ and CD4+ T-cell immune response, leading to the destruction of CD105-expressing tumor cells and tumor vasculature, thereby exerting anti-tumor and anti-angiogenic effects. LLO-CD105A has been investigated preclinically for the treatment of breast cancer and renal cell carcinoma (RCC).
