Overview

LM-305 is a novel antibody-drug conjugate (ADC) developed to target G protein-coupled receptor class C group 5 member D (GPRC5D), an orphan GPCR highly expressed in multiple myeloma cells but with limited expression in normal tissues. The drug consists of an anti-GPRC5D monoclonal antibody linked via a protease-cleavable linker to the cytotoxic payload monomethyl auristatin E (MMAE). Upon binding to GPRC5D on malignant plasma cells, the ADC is internalized and releases MMAE intracellularly, leading to microtubule disruption, mitotic arrest, and apoptosis. LM-305 was originally developed by LaNova Medicines and has been licensed globally to AstraZeneca for further development and commercialization. It is the first GPRC5D-targeting ADC to enter clinical trials for relapsed or refractory multiple myeloma[2][3][4][5][7].
