Overview

LM-364 is a Nectin-4-targeting antibody-drug conjugate (ADC) being developed by LaNova Medicines for the treatment of advanced solid tumors. It consists of a humanized monoclonal antibody specific for Nectin-4 (Poliovirus receptor-related protein 4) conjugated to a potent topoisomerase I inhibitor payload via a proprietary cleavable linker. Nectin-4 is a cell surface protein that is highly expressed in various cancers, including urothelial, breast, and lung cancers, while maintaining low expression in normal tissues. The mechanism of action involves the binding of the ADC to Nectin-4 on the tumor cell surface, followed by internalization and the subsequent release of the cytotoxic payload, which inhibits DNA topoisomerase I, leading to DNA damage and apoptosis. LM-364 is currently being evaluated in a Phase I/II clinical trial (NCT06414330) as a single agent and in combination with the anti-PD-1 antibody tislelizumab in patients with advanced solid tumors.
