Lm-GUCY2C is an investigational cancer vaccine consisting of a live-attenuated strain of *Listeria monocytogenes* (Lm) engineered to express the human tumor-associated antigen guanylyl cyclase C (GUCY2C). Developed by researchers at Thomas Jefferson University, the vaccine leverages the natural ability of *Listeria* to enter antigen-presenting cells and stimulate a potent CD8+ T-cell response against cells expressing the target antigen. GUCY2C is a membrane-bound cyclase normally restricted to the apical surface of intestinal epithelial cells but is frequently overexpressed in colorectal and small bowel adenocarcinomas, making it a viable target for immunotherapy. In clinical settings, Lm-GUCY2C is typically evaluated as a booster component in a heterologous prime-boost vaccination strategy, following a priming dose with an adenoviral vector (Ad5.F35-hGUCY2C-PADRE), to maximize the induction of tumor-specific immunity in patients with advanced gastrointestinal cancers.
