LM-IL-2Kb is a preclinical recombinant *Listeria monocytogenes* (Lm) vaccine construct engineered to co-express Interleukin-2 (IL-2) and the murine MHC class I molecule H-2Kb. Developed primarily as a research tool in the laboratory of Yvonne Paterson at the University of Pennsylvania, this construct leverages the natural ability of attenuated *Listeria* to infect antigen-presenting cells (APCs) and escape the phagosome into the cytosol. By co-expressing IL-2, the vaccine aims to provide local paracrine stimulation to enhance the activation and proliferation of T cells within the tumor microenvironment. The inclusion of the H-2Kb molecule facilitates the presentation of specific epitopes to CD8+ T cells in H-2Kb-positive mouse models, serving as a proof-of-concept for strategies to overcome immune evasion and improve the magnitude of anti-tumor immune responses.
