**Lm-LLO-E6 vaccine** is an investigational therapeutic cancer vaccine based on an attenuated Listeria monocytogenes (Lm) bacterial vector engineered to express a fusion protein of a truncated non-hemolytic version of listeriolysin O (LLO) and the E6 oncoprotein from human papillomavirus 16 (HPV16)[3][4]. The LLO segment serves as an immunogenic adjuvant, aiming to enhance immune responses against the E6 antigen, a driver of oncogenesis in HPV-associated cancers. Preclinical studies and mouse models have shown that the vaccine elicits strong tumor-specific cytotoxic T lymphocyte (CTL) responses, reduces tumor growth, and can synergize with immune checkpoint inhibitors such as anti–PD-L1 monoclonal antibodies[3][4][5]. The vaccine is primarily being developed for HPV16-positive cancers, including cervical cancer and HPV-associated lung cancer.
